Ophtalmologie expérimentale.

La section d'ophtalmologie expérimentale est un centre de recherche et d'enseignement qui a pour principal objectif d'optimiser le traitement notamment des maladies rétiniennes comme la DMLA et la rétinopathie diabétique. Notre centre d'intérêt principal est la transplantation cellulaire, la thérapie génique et la science des biomatériaux, aidé par le financement de l'Union Européenne, (FP7, TargetAMD).
Nous développons une approche pour traiter la DMLA exsudative avec la production du PEDF par des cellules modifiées génétiquement. Ces cellules transfectées permettront, une fois retransplantées sous la rétine, de synthétiser du PEDF, permettant d'inhiber la molécule angiogénique VEGF responsable de la DMLA. Nous utilisons un système de transfection non virale, et pouvons transfecter des cellules de l'épithelium pigmentaire avec un haut taux d'efficacité. Le gène est intégré de manière stable, permettant d'obtenir une production prolongée de PEDF.
Nous analysons également des biomatériaux prometteurs qui permettraient de transplanter avec une biocompatibilité optimale et afin de désigner de nouveaux dispositifs à libération prolongée. Ils sont analysés pour leurs propriétés, leur transparence, et leur maniabilité afin de déterminer leur biocompatibilité tant in vitro et qu'in vivo. Différentes cellules sont également analysées sur ces matériaux dans des études fonctionnelles et pour poursuivre l'analyse in vivo nous avons une animalerie parfaitement équipée et permettant de réaliser des examens complémentaires (OCT ou ERG) ainsi que de la chirurgie en direct.
