L'étude de l'anatomie fonctionnelle des processus cognitifs supérieurs
Les recherches de l'unité de neuroimagerie psychiatrique sont axées sur l'étude de l'anatomie fonctionnelle des processus cognitifs supérieurs (l'acte moteur volontaire, l'attention et la mémoire),
ainsi que sur la caractérisation du dysfonctionnement cérébral dans des maladies neuropsychiatriques fréquentes: psychoses, dépression et démence. Ces recherches comprennent deux thèmes principaux :
1) Anatomie fonctionnelle des processus cognitifs supérieurs. Ce thème vise à étudier les relations entre la perception et l'action, afin d'approfondir les mécanismes du dysfonctionnement du comportement humain. Nous utilisons l'imagerie par résonance magnétique (IRMf) et l'électroencéphalographie (EEG) pour étudier l'intégration des signaux sensoriels (visuels, somatosensitifs et auditifs) et les processus planificateurs de l'acte moteur volontaire. Notre objectif est de tester la part des processus cognitifs perceptuels et moteurs dans les troubles neuropsychiatriques. Pour prendre l'exemple de troubles perceptuels comme les hallucinations chez les patients schizophrènes, la question consiste à déterminer si elles résultent uniquement d'une intégration anormale des signaux sensoriels, ou si elles associent un modèle interne inadéquat de l'action. Trois projets font partie de ce thème de recherche: - Les réseaux neuronaux de l'acte moteur volontaire dans la schizophrénie : liens entre perception et action. - Le substrat anatomo-fonctionnel de l'attention visuo-spatiale chez les sujets normaux et les patients schizophrènes. - L'identification d'un dysfonctionnement précoce des circuits cortico-corticaux dans le déclin cognitif léger (Mild Cognitive Impairment) et la maladie d'Alzheimer. En collaboration avec le Pr. Giannakopoulos, Service de Psychogériatrie, HUG. 2) Métabolisme et neurotransmission cérébrale dans les maladies neuropsychiatriques. Dans certaines maladies neuropsychiatriques comme la schizophrénie et la dépression, l'hypothèse d'une régulation anormale de différents systèmes de neurotransmetteurs a été avancée. Nous développons une double approche pour étudier le métabolisme cérébral et les anomalies de la neurotransmission dans les maladies neuropsychiatriques : a) En recherche fondamentale, nous utilisons chez le rat une sonde intracérébrale qui détecte les particules beta+ (approche similaire à la TEP), et nous étudions le métabolisme cérébral et les modifications de la densité des récepteurs sérotoninergiques dans différentes conditions pharmacologiques expérimentales (par exemple, la modification des récepteurs après traitement avec des inhibiteurs de recapture de la sérotonine). b) En recherche appliquée, nous réalisons des mesures cinétiques en TEP et étudions la densité des récepteurs afin de rechercher des anomalies éventuelles spécifiques de certains tableaux neuropsychiatriques. Trois projets constituent ce thème: - Quantification in vivo des récepteurs 5HT1A et leur modulation après traitement avec des inhibiteurs de recapture de la sérotonine. - Quantification paramétrique de la densité des récepteurs gabaergiques en PET et SPECT. - Métabolisme cérébral chez le rat: mesures cinétiques in vivo et comparaison avec la méthode autoradiographique.
