Ciblage tumoral par anticorps, peptides et nucléotides radiomarqués

Ciblage tumoral par anticorps, peptides et nucléotides radiomarqués à but diagnostique et thérapeutique. Il s'agit de projets de recherche visant une application clinique, initiés généralement par une recherche translationnelle in vitro et in vivo.

Au niveau clinique, la radioimmunothérapie (RIT) du lymphome non-Hodgkin (NHL) représente une première application fructueuse de ce type de recherche avec 2 de ces traitements ayant été récemment approuvé aux Etats Unis par la "food and drug administration " (FDA). La radioimmunothérapie représente aujourd'hui une nouvelle modalité thérapeutique dans cette maladie. Nous avons été actif dans ce domaine avec une recherche dans d'autres cancers et avons participé récemment aux études phases III de RIT du lymphome. Nous poursuivons cette activité de recherche dans le domaine des anticorps anti-lymphome au niveau pré-clinique et clinique.

Comme 2ème pôle d'intérêt, nous développons un nouvel outil de ciblage, la radio-iododeoxyuridine, un nucléotide analogue de la thymidine. Ce projet bénéficie d'un soutien de la part du Fonds National Suisse de la Recherche Scientifique. Il comprend lżétude des processus biologiques impliqués dans la division cellulaire tumorale, notamment la phase de synthèse d'ADN. En modifiant la synthèse de thymidine, nous avons pu obtenir une incorporation augmentée de la radio-iododeoxyuridine dans l'ADN nouvellement synthétisée. Les premières approches thérapeutiques se font actuellement dans un modèle animal. Une étude clinique est en cours ayant pour but d'étudier, chez le patient, l'incorporation de la radio-iododéoxyuridine dans des cancers cérébraux en fonction de la modulation du cycle cellulaire qui a été testé préalablement avec succès in vitro et in vivo.

Au niveau des radio-peptides, spécifiques pour certains récepteurs exprimés par des cancers, nous avons fait une première étude chez le patient avec un nouvel analogue de la Neurotensine dans le cadre dżun projet Européen impliquant différents centres de recherche internationaux. Un nouveau projet d'étude clinique en collaboration avec le Professeur Helmut Maecke, Bâle, est actuellement en prépartation. Il s'agit d'un nouvel analogue radiomarqué de la Somatostatine qui se caractérise par une affinité plus favorable pour différentes sous-unités du récepteur de cette hormone qui sont exprimés par différents cancers neuro-endocrins.
